Reduced γδ intraepithelial lymphocyte number and motility precede the onset of Crohn’s disease-like ileitis.

Abstract Crohn’s disease (CD) is a relapsing, remitting resulting in chronic intestinal inflammation. Recent studies of resected ileal tissue from CD patients reported loss intraepithelial lymphocytes expressing the γδ T cell receptor (γδ IELs). IELs continuously survey epithelial barrier and protect against experimental colitis; however, role initiation ileitis unknown. To this end, we profiled IEL compartment TNF ΔARE/+mice, which develop CD-like at 8 wks old, prior to during onset (4–10 wks). We observed 2.5-fold reduction frequency 5-wk-old ΔARE/+mice compared +/+(WT) littermates (p<0.001). Using intravital microscopy, reduced surveillance behavior (10.6 vs 6.4 mm/min track speed mean, p<0.0001) WT 6 wks. Analysis cleared 6-wk-old revealed number villus tips relative WT, thereby increasing proportion found crypt region. Based on several links between Paneth cells (PC), quantified PCs by lysozyme immunostaining tissue. an increased morphologically abnormal reflecting PC dysfunction consistent with pathology. These findings are supported Lyz1 expression (p=0.02). Together, these demonstrate that exhibit impaired altered distribution along crypt/villus axis onset, coincides ileitis.
